整合素亚型α5β1基因抑制乳腺癌增殖和转移的作用 被引量:10

摘要 为了探讨整合素亚型α5β1基因在乳腺癌中的作用及了解其临床意义。采用Southern杂交、Northern杂交及免疫组织化学(ABC)方法,检测38例乳腺新鲜标本和105例乳腺癌石蜡切片中整合素亚型α5β1基因的表达,结合临床资料分析其临床意义。结果显示Southern杂交良性乳腺疾病整合素亚型α5β1基因的缺失率明显低于乳腺癌组;Northern杂交中良性病例的α5β1mRNA表达的量明显高于乳腺癌组;免疫组织化学的结果亦显示病理分化较好的Ⅰ级乳腺癌组α5β1基因的表达率(54.3%)高于分化差的Ⅲ级组(11.1%,P<0.05)。因此可认为整合素亚型α5β1基因的表达与乳腺疾病的良恶性质有关(P<0.05),Northern杂交结果,腋淋巴结阴性组α5β1mRNA表达的量明显高于腋淋巴结阳性组,具有显著性差异(P<0.05)。免疫组织化学检测腋淋巴结(-)、(+)组和(++)组整合素亚型α5β1基因的阳性率分别为48.8%,47.5%,38.1%(P<0.01)。无远处转移组与有远处转移组整合素亚型α5β1基因的表达率分别为53.2%与25.0%(P<0.05)。整合素亚型α5β1基因与乳腺癌的腋淋巴结? To evaluate the expression of integrin α5β1 in primary breast cancer and relationship between the changes of α5β1 gene and the clinical pathological feature,α5β1 gene DNA and mRNA expression in 38 cases of breast disease were tested by Southern blot and Northern blot.The expression of integrin α5β1 in paraffin embedded section was examined by avidin biotin peroxidase complex(ABC) test.The allelic gene deletion rate of breast cancer and benign breast diseases was 85.7%,20.0% respectively( P < 0.05 ).The α5β1 gene mRNA expression of benign breast diseases was higher than that of breast cancer,and the gene mRNA expression was related with the axllary lymph node metastasis( P <0.05).The α5β1 protein expression rate of axllary lymph node negetive(-) group,medial(+) group and strong(++) group was 48.8%,47.5 and 38.1% respectively.There was a significant difference among three group( P <0.05).The integrin α5β1 expression rate of the cancer with or without distant metastasis was 53.2% and 25.0%( P <0.01).It seems to suggest that integrin α5β1 plays an important role in the proliferation,differentiation and distant metastasis.It shows that α5β1 may be a tumor suppressor gene.
