摘要
目的采用序列分型方法(sequenced-based typing,SBT)研究深圳市公共场所冷却塔水中军团菌基因特征。方法自深圳市公共场所中央空调冷却塔水分离52株嗜肺军团菌DNA,以PCR扩增7对管家基因flaA、pilE、asd、mip、mompS、proA、neuA,并将扩增产物测序。测序结果上传欧盟军团菌感染工作组(EWGLI)数据库进行比对、分型。结果 52株嗜肺军团菌中,Lp1型共27株,ST型别分别为ST1、ST144、ST150、ST154、ST159、ST160;Lp2~14型共25株,数据库中已知型别为ST8、ST152、ST239、ST242,未知型别共9种,分别记为a~i。结论 SBT作为嗜肺军团菌分型方法,分辨率高,结果准确,易于标准化并与国际间菌株进行比对。
Objective To evaluate the gene characteristics of Legionella p neumophila isolated from the air-condition cooling tower in public places of Shenzhen by the sequence-based typing (SBT). Methods Seven housekeeping genes includingflaA, pilE,asd,mip ,mompS,proA ,neuA of 52 strains of LegioneUapneumophila were amplified by PCR. Amplified products were sequenced and the results were compared with the database of EWGLI. Results Among 52 strains of Legionellapneumophila,the ST typing of 27 strains of Lp 1 was ST1, ST 144, ST 150,ST 154, ST 159 and ST 160, 25 strains of Lp2-14 ware ST8,ST 152,ST 239 and ST 242,and 9 strains of Lp2-14 were unknown according to the database,which were named a-i. Conclusion The sequence-based typing has been proved a discriminatory, exact, normal technique, which is easy to be standardized and used to compare the international strains of Legionella pneumophila.
