摘要
采用壳聚糖为载体,以戊二醛为交联剂,通过包埋-交联耦合固定化方法,固定巨大芽孢杆菌(Bacillusm egaterium)HN7β-半乳糖苷酶,研究固定化条件和固定化酶酶学性质。和游离酶相比,以壳聚糖为载体的固定化酶,热稳定性和酸碱稳定性提高。连续催化9批次后,固定化酶酶活仍能达到最初酶活的79%,固定化酶操作稳定性较好。
In the study of effects on immobilized Bacillus megaterium HN7 β- galactosidase under different enzyme immobilization conditions and the physicochemical properties of the immobilized enzyme, we used chitosan as a carrier, and deployed embedding- linking co- immobilization method. The immobilized enzyme with the chitosan as the carrier showed high mechanical strength. The enzyme activity reached 79% of its initial activities after nine consecutive reactions, demonstrating the good operational stability of the immobilized enzyme.
