摘要
【目的】研究闭合性脑损伤后GluR2在脑组织中的表达部位、时序性特点【方法】采用清洁级SD大鼠随机分为对照组及损伤后1 h组、2 h组、4 h组、8 h组、12 h组、24 h组、2 d组、3 d组、5 d组、7 d组,复制marmarou's落体打击脑损伤模型。免疫组化SP法检测SD大鼠脑组织GluR2的表达情况。采用Mias 2000图像分析系统采集并计算阳性细胞数和阳性细胞面积。以P〈0.05为显著性差异的水准。【结果】对照组顶叶皮层、海马等部位均有免疫反应阳性细胞表达。损伤后阳性细胞表达减少,12~24 h达最低值,顶叶皮层、海马CA4、CA3、脑底皮层阳性细胞面积、阳性细胞计数与对照组比较差别有极显著意义(P〈0.01)【结论】脑损伤后,GluR2表达逐渐减少,提示其参与颅脑损伤病理生理过程。
【Objective】 To observe the temporal patterns of glutamate receptor 2(GluR2) after closed traumatic brain injury(TBI) in rats.【Methods】 Male Sprague-Dawley rats were divided into normal control and injury groups.The rats of injury group subjected to Marmarou's closed traumatic brain injury,were then subdivided into 1 h,2 h,4 h,8 h,12 h,1 d,2 d,3 d,5 d and 7 d groups according to the time elapsed after injury.The expression of GluR2 were studied with immunohistochemistry in the brain of Sprague-Dawley rats.The number and area of positive neurons were measured by Mias 2000.A ralue of P 0.05 was considered to be the level of statistical significance.【Results】 Expression of GluR2 were observed in parietal cortex,hippocampus CA4,CA3,brain bottom cortex,in sham operation control.After injury,the positive neurons expression decreased.The number of GluR2 positive neurons reached the lowest level at 12~24 h.After brain injury,the number and area of GluR2 positive neurons increased progressively in parietal cortex,hippocampus CA4,CA3,brain bottom cortex compared with sham operation control(P 0.01).【Conclusions】 After injury,the expression of GluR2 in brain tissue was decreased progressively,it had an important role in progression of brain injury.
