砷、氟、硒联合对大鼠全胚胎的发育毒性研究 被引量:1

摘要 用体外全胚胎培养(wholeembryoculture,WEC)探讨硒、氟和砷三者以不同配比浓度对旋转培养48h的大鼠胚胎所产生的联合作用。3×3×3析因分析结果表明硒、氟、砷联合对发育呈协同毒性作用;随着三种化学物剂量的配比变化,发育毒性效应亦不同,7个低剂量联合组的协同作用主要表现为致畸性,而高剂量联合组〔(2.0μg硒+10.0μg氟+1.0μg砷)/ml培养基〕的协同作用以致死性为主。结果还提示卵黄囊胎盘结构与功能紊乱是硒、氟、砷联合致畸的重要机制之一。 Whole embryo rotated culture technique was used to investigate the toxicity of combination of selenium,fluoride and arsenic on rat embryos at day 9 5 of gestation.The result of factorial analysis (3×3×3)showed that the main effect of combinafion of selenium, fluoride and arsenic on the developmental toxicity was synergistic.The mixtures with different level of these three chemicals in combination could result in different developmental toxicity.The low level combinations mainly caused teratogenic effect,and the high level combinations(selenium 2 0μg+ fluoride 10μg+arsenic 1 0μg/ml culture media)caused lethal effect. The results suggested that the disorders of yolk sac placenta in structure and function were one of teratogenic mechanisms for the combination of selenium,fluoride and arsenic.
