摘要
应用分子生物学技术 ,将编码甲型肝炎病毒 (HAV )外壳蛋白的开放读码框架 (ORF)全序列插入杆状病毒多角体启动子下游 ,构建成重组杆状病毒 (r BHAV ) ,并在昆虫细胞 SF9中表达。实验证明 ,在该重组杆状病毒中含有完整的 HAV编码序列 (HAV ORF) ;表达的重组蛋白具有良好的抗原性和免疫原性 ;电镜检查被 r BHAV感染的 SF9细胞中有与甲肝病毒颗粒类似的球形结构 ;经氯化铯梯度离心测得该重组蛋白的浮力密度与 HAV空壳一样。
Recombinant baculoviruse was constructed by inserting a full length cDNA open reading frame( ORF) coding HAV capsid protein into the end of polyhedrin promoter and expressed in SF9 cell.The result proved that the recombinant baculoviruse contained a full length HAV ORF,and the expressed protein showed good antigenicity and immunogenicity.HAV particle like spherical structure was observed in the SF9 cells infected with the recombinant baculoviruse under electron microscope.The buoyant density of the recombinant protein detected by cesium chloride gradient centrifugation was similar to that of HAV capsid.
