摘要
建立 1种筛选和鉴定生物制品免疫活性的方法—— BSA免疫细胞化学法 CD+ 2 细胞增长试验。经预试 ,确定了胰酶的最适浓度和处理时间 ,业经处理的 PBMC温育的合适时间和药物的最适浓度 ,并对已知具有免疫增强或免疫抑制功能的若干种制剂作活性检测 ,表明本法具有操作简便、重复性好、易标准化的优点。
A new method for screening and evaluation of immunomodulator, CD + 2 cell proliferation test, was established. After the CD 2 antigens on the surface of peripheral blood mononuclear cells(PBMC) were digested with trypsin, the PBMCs were incubated at 37℃ together with immunomodulator for a appropriate time, then the CD + 2 cell proliferation rate was detected by biotin streptavidin immunocytochemistry method and used for evaluation the activity of immunomodulator. The activities of several kinds of known immunopotentiators and immunosuppressants were detected by the method, and the optimal concentractions of trypsin and the optimal time for digestion and incubating were studied. The method was simple and showed good reproducibility. It was also easy to be standardized.
