细鳞裂腹鱼同工酶组织特异性研究 被引量:13

摘要 采用聚丙烯酰胺凝胶电泳法对细鳞裂腹鱼的5种组织(心脏、肝、脾、肌肉、脑)进行了5种同工酶(ADH、LDH、MDH、MEP、POD)的初步研究.结果表明:细鳞裂腹鱼同工酶具有明显的组织特异性.ADH同工酶表达为3条酶带,肝脏的Adh-A2为优势表达带;LDH同工酶在组织中表达为5条酶带,Ldh-B4在心脏中优势表达,Ldh-A4在肌肉中优势表达;MDH同工酶共有4条酶带,脾脏活性最强;测到的MEP共有4条酶带,脑只表达不明显的2条酶带(Mep2、Mep3),活性均较低;POD同工酶在心脏中出现2条酶带,其他组织均表达为1条酶带,在心脏和脾脏中优势表达. Five isozymes expressed in 5 tissues, i.e. heart, liver, spleen, muscle and brain, of Schizothotax chongi were preliminarily investigated by polyacrylamid gel electrophoresis (PAGE). The result showed that isozymes were expressed in S. chongi with remarkable tissue-specificity. There were 3 ADH bands found in the tissues, and Adh-A2 was expressed dominantly in the liver. Five LDH bands were found in the tissues, and Ldh-B4 isozymic band was expressed dominantly in the heart and Ldh-A4 isozymie band in the muscle. Four zymograms of MDH were pigmented, and the strongest activity was pigmented in the spleen. Four MEP bands were pigmented in the tissues with weaker activity, except 2 bands (Mep2 and Mep3) in the brain tissue. POD isozyme was dominantly expressed in the heart and the spleen, with 2 bands in the heart and 1 band in other tissues.
