摘要
目的探讨踝蛋白1(Talin1)在正常肝组织、原发性肝细胞癌(HCC)组织、癌旁组织中的表达情况;探讨Talin1表达与HCC发生及其预后的关系。方法采用免疫组织化学方法 (SP法)检测正常肝组织(10例)、HCC组织(32例)及其对应癌旁组织(32例)中的Talin1表达情况。结果正常肝组织、HCC组织及癌旁组织中均有Talin1表达,其阳性表达率分别为100%、62.5%和96.9%。Talin1表达于阳性细胞胞质及胞膜上;Talin1在HCC组织中的表达显著低于癌旁组织和正常肝组织(P<0.05);而正常肝组织与癌旁组织中的Talin1表达差异无统计学意义(P>0.05);Talin1的表达与肝癌患者性别、年龄、肿瘤个数、AFP表达水平及HB-sAg无关(P>0.05),与患者术后一年无瘤生存率无关(P>0.05);而与肝癌病理分化程度、包膜完整性、门脉癌栓形成及肿瘤大小相关(P<0.05)。结论 Talin1与HCC的癌变、浸润和转移有关;Talin1可作为HCC发生、发展的预测指标之一。
Objective To detect the expression of Talin1 in the normal liver tissues,primary hepatocellular carcinoma tissues and paracarcinomatous tissues and investigate the relationship between Talin1 expression and the oncogenesis and prognosis of HCC.Methods The expression of Talin1 was detected by immunohistochemical SP method in the normal liver tissues(n=10),HCC tissues(n=32)and its paracarcinomatous tissues(n=32).Results The expression of Talin1 was observed in the normal liver tissues,HCC tissues and paracarcinomatous tissues.The positive rate of Talin1 were 100%,62.5%,96.9% respectively.The brown granules of positive cell were mainly distributed in the cytoplasm and cell membrane.The expression of Talin1 in HCC tissues was significantly lower than that in the paracarcinomatous tissues and normal liver tissues(P〈0.05),but between paracarcinomatous tissues and normal liver tissues had no significance(P〈0.05).Talin1 protein expression was not related to sex,age,tumor number,the expression of AFP and HBsAg(P〉0.05),and had no correlation with the one-year disease-free survival of patients(P〉0.05),but were significantly correlated with pathological differentiation of HCC,the integrity of tumor capsule,portal vein tumor thrombus and tumor size(P〉0.05).Conclusion Talin1 is related to the carcinogenesis,infiltration and metastasis of HCC,The level of Talin1 expression may be a valuable marker for diagnosis and prognosis of HCC.
