摘要
目的分析阴道毛滴虫(Trichomonas vaginalis,Tv)和口腔毛滴虫(Trichomonas tenax,TT)的同工谱特征及其亲缘关系,寻找同工酶谱鉴别指标。方法采用聚丙烯酰胺凝胶垂直板电泳,分别进行苹果酸脱氢酶(malate dehydrogenase,MDH)、谷氨酸脱氢酶(glutamic dehydrogenase,GLDH)、6-磷酸-葡萄糖脱氧酶(glucose-6-phosphate dehydrogenase,G6PD)、乙醇脱氢酶(alcohol dehydrogenase,ADH)和过氧化氢酶(Catalase)5种同工酶染色,比较分析2种滴虫的酶谱。结果 MDH同工酶:2种滴虫只有相对迁移率(Rm)52、852条区带相同,而Rm34、42、94、1004条区带在2种滴虫之间明显不同;GLDH同工酶:TV无显带,TT共分离出6条酶带;G6PD同工酶:TV只有Rm35、42、523条区带,而TT除了显示这3条区带外,还显示出其他迁移率更大的区带;ADH和Catalase均无区带显示。结论 MDH、GLDH和G6PD3种同工酶均具有鉴别意义。
Objective To analyze the characteristics of isozymograms of Trichomonas vaginalis ( TV) and Trichomonas tenax ( TT) and their relation in an attempt to find the identification index of their isozymo-grams. Methods Five isozymes including melate dehydrogenase ( MDH) ,glutamic dehydrogenase ( GLDH) ,glucose-6-phosphate dehydrogenase ( G6PD) ,alcohol dehydrogenase ( ADH) and catalase were stained with SDS-PAGE. Isozymograms of TV and TT were compared and analyzed. Results Two identical bands ( Rm52 and Rm85) of MDH isozymogram and 4 bands ( Rm34,Rm42,Rm94 and Rm100) of MDH isozymogram were found in TV and TT,respectively. No band and 6 band of GLDH isozymogram were identified in TV and TT,respectively. Three bands ( Rm35,Rm42 and Rm52) of G6PD isozymogram were observed in TV,and more bands of G6PD isozymogram were demonstrated in TT,in addition to the 3 bands in TV. No band of ADH and catalase isozymogram was displayed in TV and TT. Conclusion MDH,GLDH and G6PD can be used as an identification index of TV and TT isozymograms.
