吲哚美辛对A549/DDP细胞摄取^(99)Tc^m-MIBI的影响及其逆转耐药性的机制 被引量:5

摘要 目的:探讨吲哚美辛对A549/DDP摄取99Tcm的影响及其逆转耐药性的机制。方法:常规方法培养A549/DDP细胞,首先行99Tcm-MIBI摄取实验,然后用DDP和吲哚美辛干预A549/DDP细胞30min后计算99Tcm-MIBI的摄取率,再分别用RT-PCR,Westen-blot检测mdr1基因,Pg-p蛋白的表达。结果:A549/DDP细胞摄取率在90min时达到高峰(0.83%),吲哚美辛干预后99Tcm-MIBI摄取比干预前增加,达到3.79%。RT-PCR,Westen-blot结果显示吲哚美辛抑制了mdr1基因和Pg-p蛋白的表达。结论:吲哚美辛能提高A549/DDP细胞99Tcm摄取率并通过抑制mdr1和Pg-p的表达逆转肺癌耐药。99Tcm放射性摄取率可作为评价肿瘤耐药的一种方法。 Objective: To explore the mechanism of indomethacin reversal cisplatin resistance on lung cancer cells. Methods: With the conventional method,to cuhivat A549 / DDP cells. To do99Tcm -MIBI intake experiment first, then use DDP and indomethacin intervent cell . After 30mins to calculate cells 99Tern - MIBI uptake, then with RT- PCR,western- blot to detect- MDR1 gene and Pg -p protein expression. Results:A549/DDP cells, uptake of 99Tern -MIBI aehived peak in 90 rain (0. 83% ), 99 Tom -- MIBI intake of indomethacin intervention group increased than DDP intervention group significantly (3.97%) ,Rusults of RT- PCR ,and Western- blot showed indomethacin coued inhibit MDR1 genes and Pg -p protein expression. Conclution: Indomethacin can inhibit MDR1 ,Pg - p ex- pression and reversedrag drag resistance of lung cancer.
