摘要
目的探讨5-氮杂胞苷(5-aza-CR)对HL-60细胞中XIAP和Caspase-9表达水平的影响以及XIAP与Caspase-9表达的相关性,初步了解XIAP抗凋亡的途径。方法培养HL-60细胞,用不同浓度5-aza-CR干预,流式细胞术检测干预前后XIAP和Caspase-9的表达水平。结果 5-aza-CR干预HL-60细胞后早期凋亡率在5-aza-CR各浓度组均随干预时间的延长而增加,其中10.0μmol/L组干预4h后凋亡率达(34.26±3.15)%;XIAP表达水平在5-aza-CR各浓度组均随干预时间的延长而下降,10.0μmol/L组干预4h后降至76.22±2.84;Caspase-9表达水平在5-aza-CR各浓度组均随干预时间的延长而升高,10.0μmol/L组干预4h后升至75.45±4.64;XIAP和Caspase-9表达水平呈负相关。结论 HL-60细胞中XIAP抗凋亡途径可能与抑制Caspase-9表达相关。
Objective To detect influence of 5-azacytidine on expression level of XIAP and caspase-9 in HL-60 cells,and analyze the relationship between XIAP and caspase-9.Methods HL-60 cells was co-cultured with 5-azacytidine in different concentrations and the cells was collected in different time points.FCM was used to detect expression level of XIAP and caspase-9.Results Early apoptosis rate of HL-60 cells after co-cultured with 5-azacytidine was increased in concentration and time dependent manner.It was(34.26±3.15)% in 10.0μmol/L group after 4 hours.Expression level of XIAP was decreased by 5-azacytidine in concentration and time dependent manner in HL-60 cells.It was 76.22±2.84 in 10.0μmol/L group after 4 hours.Expression level of caspase-9 was increased by 5-azacytidine in concentration and time dependent manner in HL-60 cells.It was 75.45±4.64 in 10.0μmol/L group after 4 hours.XIAP was negative correlation with caspase-9.Conclusion The mechanism of XIAP anti-apoptosis is correlated with caspase-9.
