食蟹猴MHC Ⅰ类A型新等位基因序列分析

Sequence Analysis of Macaca fascicularis's New MHC Ⅰ-A Alleles

摘要 采用自行设计的MHC I类A型基因的引物从10只食蟹猴扩增得到11个3000bp左右MHC等位基因。通过在Blast比对之后,证实有7个Mafa-A等位基因,4个Mafa-AG等位基因,新的等位基因序列已递交GenBank。由于每个个体可以检测出多个不同的Mafa-A等位基因,这说明食蟹猴Mafa-A基因经过多次复制,并且推测Mafa-AG是Mafa-A衍生物。同时,研究了等位基因在个体中的分布和系统进化树分析。 Elevent＇s MHC class Ⅰ alleles around 3 000bp have been identified from 10 Macaca fascicularis.BLAST analysis showed that the novel alleles include 7 Mafa-A alleles and 4 Mafa-AG alleles,which had been submitted to GenBank.Different Mafa-A alleles have been identified in each sample,it can be inferred that Mafa-A alleles have been duplicated and Mafa-AG alleles are derivative of Mafa-A alleles.Based on the new alleles.The alleles distribution among the samples and the phylogenetic tree were analyzed.
