摘要
目的探讨来曲唑对大鼠子宫内膜异位症(EM)模型异位病灶中血管内皮生长因子(VEGF)及微血管密度(MVD)表达的影响,及其治疗EM的作用机制。方法行自体移植建立EM大鼠模型,将建模成功大鼠随机分为来曲唑组及盐水对照组,比较口服来曲唑或盐水前后EM大鼠异位病灶体积改变,并采用免疫组化法检测各组异位内膜VEGF及MVD的表达变化。结果来曲唑组异位病灶体积明显缩小或萎缩,与对照组比较差异有统计学意义(P<0.05)。来曲唑治疗后VEGF、MVD在异位内膜中表达降低(P均<0.05)。结论来曲唑对大鼠异位囊肿的体积影响明显,短时间内就能起到明显效果。可能通过降调VEGF、MVD在异位内膜组织中的表达而发挥治疗作用。
Objective To study the different expression patterns of vascular endothelial growth factor(VEGF)and micro-vessel density(MVD)in tissues of endometriosis in rat models after letrozole therapy,and to explore the underlying mechanism of letrozole treatment for endometriosis(EM).Methods Rat models were established by autologous transplantation of uterine tissues to an ectopic site beside the uterines.Then the successful rat models were randomly divided into the NS control group and the Letrozole(p.o.)treated groups.To compare the difference of ectopic lesion volumes after the treatment,an immunohistochemical method was used to detect VEGF and MVD expressions in tissues of endometriosis in the rat models.Results The volume of endometriotic tissues in the letrozole groups decreased compared with the control group(P0.05).The inhibitory effect of the drug significantly increased with the duration of treatment.Expressions of VEGF and MVD in tissues of endometriosis decreased after using letrozole.Compared with the control group,the differences were significant(P0.05).Conclusions The volume of the ectopic focus in rat models was obviously changed after letrozole treatment,and a significant effect appeared in a short time.Letrozole can treat EM through down-regulating expressions of VEGF and MVD in ectopic tissues.
