15份葡萄种质遗传多样性的SSR分析 被引量:25

摘要 本研究利用SSR标记对15份葡萄种质进行了遗传多样性分析。从30对SSR引物中筛选出11对多态性引物,共扩增出107条带。每对引物可检测到等位位点数为4~22,多态率为75%~100%。根据SSR扩增结果,利用NTSYSpc2.10e软件进行Jaccard相似性系数分析,15份葡萄种质的遗传相似系数在0.20~0.98,平均遗传相似系数为0.398。UPGMA聚类分析结果表明,在遗传相似系数0.29处,15份葡萄材料可分为2大类群。第一类包含7份山葡萄资源,2个山欧杂交品种,第二类包含3个欧亚种品种、2个欧美杂种品种和1个美洲杂种品种。由此可见,山葡萄与欧亚种、美洲杂种的亲缘关系较远,欧亚种与美洲种之间亲缘关系较近。此外,引物Vmc9a2.1、VMC4F3、VMC4A1、Scu14vv分别在欧亚种‘无核白’、‘山葡萄雄株’(雄1-2,雄1-3)、山葡萄‘左山二’、山葡萄‘双丰’扩增出一条特性条带,这为利用SSR标记鉴定葡萄品种或品系提供了基础数据。 In this paper,we conducted genetic analysis of 15 grape germplasms by using SSR markers.The results showed that 107 bands were amplified by using 11 pairs of polymorphic primers screened from 30 pairs.No.of allelic loci detected by each pair of primers ranged from 4 to 22,with the polymorphic rate of 75%~100%.Jaccard similarity coefficient analysis by NTSYSpc2.10e software based on SSR amplification results demonstrated that the genetic similarity coefficient was 0.20~0.98,with the average genetic similarity coefficient of 0.398.The UPGMA clustering analysis showed that the 15 grape germplasms could be divided into two major groups on similarity coefficient level of 0.29,among which seven Vitis amuensis varieties and two hybrid varieties derived from(V.amurensis×V.vinifera)×V.amurensis were clustered in the same group and the second group included three V.vinifera varieties,two Euro-American hybrid varieties and one American hybrid variety.The results showed that the relationships between V.amurensis and V.vinifera,V.amurensis and Euro-American hybrid was relatively farther,while that between V.vinifera and Euro-American hybrid,V.vinifera and American hybrids was closer.In addition,only one specific band was amplified from Thompson seedless,Vitis amurensis Male 1-2 and male 1-3,Zuoshaner,Shuangfeng by using primer Vmc9a2.1,VMC4F3,VMC4A1 and Scu14vv,respectively,which provide basic data for identification of grape varieties or lines by using SSR markers.
