摘要
目的 探讨雌激素受体阳性(ER+)乳腺癌细胞MCF-7和雌激素受体阴性(ER-)乳腺癌细胞MDA-MB-231及MDA-MB-435s中E1AF,MMP-1和MMP-9表达情况及相关性.方法 应用免疫细胞化学技术检测3种不同乳腺癌细胞中MMP-9的表达,采用RT-PCR方法检测细胞株中E1AF,MMP-1及其mRNA表达.结果 ER(-)乳腺癌细胞MDA-MB-231中E1AF基因、MMP-1及其mRNA的表达水平与MMP-9的表达高于ER(+)乳腺癌细胞MCF-7和ER(-)乳腺癌细胞MDA-MB-435s.结论 乳腺癌细胞中E1AF的表达与MMP-1,MMP-9的表达存在相关性,E1AF可能是乳腺癌侵袭转移的重要因子.
Objective To explore the relationship among the expression of E1AF,MMP-I and MMP-9 in the estrogen receptor positive(ER + ) MCF-7 and negative(ER - ) MDA-MB-231, MDA-MB-435s human breast cancer cell lines. Method The EI AF gene, MMP-1 and mRNA of human breast cancer cell lines were detected using RT-PCR technique and MMP-9 using immunocytochemistry. Results The E1AF gene, MMP-1 and mRNA and MMP-9 were at high level in the estrogen receptor negative( ER - ) MDA-MB-231 cells,but lower in estrogen receptor positive( ER + ) MCF-7 and negative( ER - ) MDA-MB-435s cells. Conclusion The expression of E1AF was correlated with the level of MMP-1 and MMP-9. Maybe E1AF is an important factor in the invasion and metastasis of human breast cancer.
