不同浓度葡萄糖对心肌细胞损伤的研究

摘要 拟证实高浓度葡萄糖环境可致心肌细胞脂代谢紊乱、亦可直接引起心肌细胞炎症损伤,以不同浓度葡萄糖培养液为干预因素进行实验.以脂代谢重要酶—脂蛋白脂酶的表达为中心观察心肌细胞脂代谢改变;选择常见炎症介质TNF-α为标志观察心肌细胞的炎症反应.结果显示,在50mmol/L葡萄糖作用下正常心肌细胞TNF-αmRNA表达(0.58±0.03)明显高于20mmol/L(0.27±0.02)、30mmol/L(0.29±0.05)葡萄糖作用的正常心肌细胞,统计学比较有差异(P<0.05),而后2组比较无统计学差异(P>0.05).表明,高浓度葡萄糖环境中,心肌细胞脂蛋白脂酶表达上调及脂蛋白脂酶活性增高,细胞内脂质堆积,出现脂代谢紊乱;心肌细胞TNF-αmRNA表达上调,引起炎症反应,这2方面的改变均可导致心肌细胞病理性损伤. Our objective was to demonstrate that high concentrations of glucose could not only induce the lipid metabolic turbulence but also bring inflammatory trauma to the cardiomyocytes.Experiments were made on different concentrations of glucose.The changes of lipid metabolism of cardiomyocytes were observed round the center of LPL as it was an important enzyme of lipid metabolism.The inflammatory reaction of cardiomyocytes was observed by choosing the common inflammatory factor TNF-α.The results showed that the expression of TNF-αmRNA on the normal cardiomyocytes treated with 50mmol/L glucose (0.58±0.03)was obviously higher than those when treated with 20 mmol/L glucose(0.27±0.02) and 30 mmol/L glucose(0.29±0.05).In statistics,the difference was (P〈0.05) but the latter two had no such a statistic difference.Hence,our conclusion was that the expression of LPL was increased and its activity was enhanced.Lipid was accumulated,resulting in its metabolic turbulence.The expression of TNF-αmRNA was increased,thus causing inflammatory reactions.Both changes could induce pathologic lesion of cardiomyocytes.
