凋亡抑制基因Survivin和Pten在胆囊癌中的表达及相关分析

摘要 ①目的通过研究Survivin与Pten在胆囊癌中的表达,探讨二者的相关性。②方法采用免疫组织化学方法检测Survivin与Pten在51例胆囊癌标本和13例慢性胆囊炎及26例正常的胆囊组织标本中的表达。③结果Survivin在51例胆囊癌中呈阳性表达的有40例,表达率为80.4%,而在13例慢性胆囊炎组织和26例正常的胆囊组织中均无阳性表达,两者之间的差异具有显著性(P<0.01)。Pten在胆囊癌中的阳性表达率为29.4%,在非肿瘤性胆囊组织(慢性胆囊炎、正常的胆囊组织)中的阳性表达率100%,两者差异显著(P<0.01)。Survivin和Pten的表达与患者的性别、年龄、胆囊癌的临床分期、淋巴结转移无关。Survivin和Pten在胆囊癌的表达呈负相关性(r=-0.499,P=0.000)。④结论Pten对胆囊细胞的增生起抑制作用,Survivin对胆囊细胞的凋亡起抑制作用,Survivin在胆囊癌中的高表达和Pten在胆囊癌中的失表达可能与胆囊癌的发生发展有关,检测Survivin和Pten的表达将有助于对胆囊癌的诊断。 Objective To investigate the expression of Survivin and Pten in gallbladder carcinoma and study their relationship.Methods Expression of Survivin and Pten was detected by immunohistochemical technique in 51 cases of gallbladder carcinoma,13 cases of chronic cholecystitis and 26 cases of normal gallbladder tissue.Results Positive expression rate of Survivin was 80.4% in gallbladder carcinomas(40/51),and which was 0 in 13 cases of cholecystitis and 26 cases of normal gallbladder tissue,while positive expression rate of Pten in gallbladder carcinomas was 29.4%,and which was 1000% in chronic cholecystitis and normal gallbladder tissue,and there were significant difference between two groups(P0.01).Expressions of Survivin and Pten were no correlation with patient age,sex,clinical stage of gallbladder carcinoma and lymph metastasis.There was negative relation between Survivin expression and Pten expression(r=-0.499,P=0.000).Conclusion Pten played an inhibitive roles in proliferation of gallbladder cells,but Survivin did in apoptosis.High expression of Survivin and Low expression of Pten may be correlation with the progression of gallbladder carcinoma.It will be help for diagnosis of gallbladder carcinoma to detect expressions of Survivin and Pten.
