摘要
目的:探讨共轭亚油酸对妊娠期糖尿病大鼠胎盘脂联素基因表达水平的影响。方法:育龄雌性SD大鼠受孕后随机分为正常妊娠组,妊娠期糖尿病组,共轭亚油酸治疗组。腹腔内注射45mg/kg链尿佐菌素,建立妊娠期糖尿病模型。注射3天后用共轭亚油酸3.0g/(kg.d)灌胃至孕19天,建立共轭亚油酸治疗组模型。用葡萄糖氧化酶法测定血糖;采用RT-PCR技术检测胎盘脂联素基因mRNA的表达水平,并进行半定量分析。结果:(1)孕前3组血糖差异无统计学意义;孕3天后妊娠期糖尿病组、共轭亚油酸治疗组血糖水平显著高于正常妊娠组,妊娠期糖尿病组与共轭亚油酸治疗组差异无统计学意义;孕19天后妊娠期糖尿病组血糖水平显著高于正常妊娠组,共轭亚油酸治疗组血糖水平显著低于妊娠期糖尿病组,但仍高于正常妊娠组;(2)胎盘组织中有脂联素基因表达;(3)妊娠期糖尿病组胎盘脂联素基因mRNA的表达水平显著低于正常妊娠组;共轭亚油酸治疗组胎盘脂联素基因mRNA的表达水平显著高于妊娠期糖尿病组,但仍低于正常妊娠组。结论:(1)共轭亚油酸可降低妊娠期糖尿病大鼠血糖水平;(2)胎盘部位有脂联素基因表达,妊娠期糖尿病病理状态可影响脂联素基因在胎盘的表达水平;(3)共轭亚油酸有上调脂联素基因表达水平的作用,可改善妊娠期糖尿病大鼠的糖脂代谢。
Objective:To study the effects of conjugated linoleic acid(CLA) on the expression level of adiponectin(APN) mRNA in placenta of gestational diabetes mellitus(GDM) rats.Methods:Pregnant reproduction age SD rats were randomly devided into three groups which included normal pregnancy group,GDM control group,diabetic CLA treatment group[i.g.CLA 3.0g/(kg·d)].Abdominal injection of Streptozotocin(STZ) 45mg/kg induced GDM in SD rats.The treatment of CLA to GDM rats from injecting STZ to pregnant 19 days.Glycemia was detected by glucose oxidase method.The expression level of APN mRNA in placenta was evaluated by RT-PCR.Results:(1)Progestationally,the glycemia of three groups had no significant difference;3 days after prengnancy,the glycemia of GDM group and CLA treatment group was significantly higher than that in normal pregnancy group,but GDM group and CLA treatment group had no significant difference;19 days after prengnancy,the glycemia of GDM group was significantly higher than that in normal pregnancy group and the glycemia of CLA treatment group was significantly lower than that in GDM group,but was still higher than that in normal pregnancy group.(2)The APN mRNA was expressed in placenta.(3)The expression level of APN mRNA in GDM group was significantly lower than that in normal pregnancy group;the expression level of APN mRNA in CLA treatment group was significantly higher than that in GDM group but was still lower than that in normal pregnancy group.Conclusion:(1)CLA reduced the level of glycemia of GDM rats.(2)There was expression of APN mRNA in placenta.The expression level of APN mRNA in placenta may be influenced in the condition of GDM.(3)CLA increased the expression level of APN mRNA in placenta and improved the glucose and lipid metabolism of GDM rats.
