摘要
探讨CD122分子表达水平与非肥胖型糖尿病(NOD)小鼠生育力低下的关系。采用磁珠亲和细胞分选术分离小鼠子宫NK细胞,采用流式细胞术检测NK细胞表达CD122分子的情况,比较与C57BL/6雄鼠同笼而受孕的NOD小鼠(NOD×C57BL/6)与对照组BALB/c×C57BL/6小鼠流产率的差异以及CD122表达水平的差异。结果显示,NOD×C57BL/6小鼠孕12.5 d胚胎吸收率显著高于BALB/c×C57BL/6对照组(分别为25.7%和2.4%,P<0.01),而其子宫NK细胞CD122阳性率显著低于对照组(分别为28.1%和51.4%,P<0.01)。进一步检测发现,这些细胞不表达CD3和CD4分子。过继输注BALB/c×C57BL/6孕鼠子宫NK细胞可降低NOD×C57BL/6小鼠胚胎丢失率,而注射中和抗体阻断CD122可升高NOD×C57BL/6小鼠和BALB/c×C57BL/6小鼠胚胎丢失率。这些结果提示,NOD×C57BL/6小鼠自发性胚胎丢失率增高可能与缺乏足够的CD122阳性子宫NK细胞有关。
To investigate the relationship between the status of CD122 expression and the sub-fertility in non-obese diabetic (NOD) mice,magnetic affinity cell sorting was performed to isolate uterine natural killer(NK) cells,and flow cytometry was used to detect the status of CD122 expression in NK cells.The resorption rate of embryos and the proportion of NK cells producing CD122 molecule were compared between NOD×C57BL/6 and BALB/c×C57BL/6 mice at day 12.5 of gestation.It was demonstrated that the resorption rate in NOD×C57BL/6 mice was significantly higher than BALB/c×C57BL/6 mice(25.7 versus 2.4%,P0.01),whereas the percentage of CD122~+ cells in NK cell population in NOD×C57BL/6 mice were significantly lower than BALB/c×C57BL/6 mice(28.1%versus 51.4%,P0.01).Further detection showed that no CD3 or CD4 molecules were produced by these cells.The adoptive transfer of uterine NK cells from BALB/c×C57BL/6 mice could decrease the percentage of embryo loss in NOD×C57BL/6 mice,whereas neutralizing antibody against CD122 increased embryo resorption rate in both NOD×C57BL/6 and BALB/c×C57BL/6 mice.These results suggest that sufficiency of CD122~+ NK cells in NOD×C57BL/6 mice may be correlated with its increased occurrence of embryo resorption.
