摘要
目的了解HyTK基因转移联合Ganciclovir(GCV)对人膀胱癌细胞的杀伤作用。方法采用570型粘附式细胞激光定量分析筛选仪测定GCV作用下人膀胱癌细胞株EJ细胞、转导HyTK基因的EJ细胞(EJ/TK)DNA含量的变化。结果实验组EJ/TK细胞DNA平均荧光值159208±18070,明显低于对照组204717±22841(P<0001);而EJ细胞DNA平均荧光值实验组为224606±234.66,对照组为223297±252.08,两组间差异无显著性(P>05)。结论GCV作用能明显降低转导HyTK基因EJ细胞(EJ/TK)的DNA含量。
Obejective To elucidate the ganciclovir effect on human bladder carcinoma cells. Methods The DNA fluorescence intensity of human bladder carcinoma cell line EJ cells and HyTK gene transduced EJ cells (EJ/TK) was measured by adherent cell analysis and sorting cytometer (ACAS570).In experimental group,EJ/TK cells and EJ cells were cultured respectively and treated with GCV of 100 μmol/L for 48h,while GCV was absent in controls.Cells were stained by propidium iodide (PI). Results The average DNA fluorescence value of EJ/TK7 cells was 1 592.08±180.70 and that of EJ cells was 2 246.06±234.66 in the experimental group;while the average DNA fluorescence value of EJ/TK cells was 2 047.17±228.41 and that of EJ cells was 2 232.97±252.08 in controls.There was no significant difference between experimental and control group of EJ cells ( P >0.5);Comparing with the control,thd average DNA fluorescence value of EJ/TK cells after exposure to GCV decreased significantly ( P <0.001). Conclusions GCV could obviously reduce the DNA content of HyTK gene transduced EJ cells and suggest HyTK gene transfer in conjunction with GCV have therapeutical effect on human bladder cancer.
