摘要
目的观察缺氧刺激对大鼠骨髓间充质干细胞(rBMSCs)骨向诱导分化过程中碱性磷酸酶(ALP)活性的影响。方法 rBMSCs骨向诱导后按不同氧浓度及不同作用时间分组,检测不同作用时间的缺氧对rBMSCs骨向诱导分化过程中ALP活性的影响。结果缺氧培养对rBMSCs骨向诱导分化中ALP活性的影响除12 h略有降低但无显著性差异外,其余各时间段均有明显抑制作用且有显著性差异(P均<0.05)。结论缺氧培养对rBMSCs骨向诱导分化过程中ALP活性有明显抑制作用,提示2%缺氧对rBMSCs骨向分化的成骨效应有抑制作用,进而促使骨改建平衡向破骨方向倾斜。
Objective It is to observe the influence of hypoxia stimulus on alkaline phosphatase(ALP) activity in rat bone marrow mesenchyme stem cells(rBMSCs) in the course of differentiating into osteoblast.Methods The hypoxia model of rBMSCs was divided into groups according to different oxygen density and different time to detect the influence of different time hypoxia on ALP activity.Results The influence of hypoxia cultivation on ALP activity of rBMSCs in the course of differentiating into osteoblast at 12 h was slightly lowered but had no significant difference,and that at the other time had all obvious inhibitory action and had significant difference(all P0.05).Conclusion Hypoxia cultivation has obvious inhibitory action on ALP activity of rBMSCs in the course of differentiating into osteoblast,which hints that 2% hypoxia has inhibitory action on ossification action of rBMSCs in the course of differentiating into osteoblast and then promotes bone remodeling equilibrium inclines bone fracture.
