摘要
目的探讨华蟾素(cinobufagin)对人骨肉瘤细胞株MG-63的促凋亡作用。方法采用MTT比色法、透射电镜观察、原位末端标记tunnel法、流式细胞仪检测等方法,观察和检测华蟾素对MG-63的促凋亡作用。结果华蟾素对MG-63骨肉瘤促凋亡作用明显,且呈现时间和浓度依赖性。华蟾素将MG-63细胞阻断在S期,可诱导骨肉瘤细胞凋亡。与甲氨蝶呤对照组相比,不同浓度下华蟾素对MG-63细胞促凋亡作用更显著。结论华蟾素对MG-63骨肉瘤促凋亡明显。华蟾素可用于骨肉瘤治疗。
Objective To study the cytotoxicity of cinobufotalin on MG-63 osteogenic sarcoma cells and its mechanism of action. Methods The cytotoxicity of cinobufotalin on MG-63 osteogenic sarcoma cells was observed and tested through MTT cotorimetry, electronic microscope, in situ end-labeling tunnel method, and flow cytometry test, ect. Results Cinobufagin has significant cytotoxicity on MG-63 osteogenic sarcoma cells, and demonstrated time and concentration dependence. Cinobufagin blocked MG-63 osteogenic sarcoma cells at stage S, and induced the death of osteogenic cells. Compared with the methotrexate group, cinobufotalin has better pro-apoptic effect on MG-63 cells in different concentration. Conclusions Cinobufotalin has strong pro-apoptic effect on MG-63 osteogenic sarcoma cells. It can be used to treat osteosarcoma.
