摘要
目的研究法舒地尔对急性氧化乐果中毒大鼠心肌损害的保护作用。方法将30只雄性SD大鼠随机分为对照组、阿托品组和法舒地尔组(每组n=10),采用腹腔注射氧化乐果制备急性有机磷农药中毒模型,染毒后给予阿托品或阿托品+法舒地尔的不同治疗,观察各组大鼠存活状况、心肌病理学变化和心肌细胞凋亡情况。结果与对照组比较,阿托品组和法舒地尔组大鼠心肌病理损害明显,心肌细胞凋亡显著增加(P<0.01);但与阿托品组比较,法舒地尔组存活率较高;心肌病理损害较轻,心肌细胞凋亡显著减少(P<0.01)。结论法舒地尔对急性氧化乐果中毒大鼠的心肌损害有保护作用。
Objective To investigate the protective effect of fasudil on the myocardial injury in rats with acute omethoate poisoning.Method 30 male Sprague-Dawley(SD) rats were randomized into the control group,the atropine group and atropine plus fasudil group(Fasudil group)(n=10 each).The omethoate poisoning model was established by peritoneal injection of omethoate.After poisoning,the rats were treated with atropine or atropine plus fasudil,and the survival rate was observed.The pathological changes of the cardiac tissue was observed by H-E staining and the myocardiocyte apoptosis was determine by TUNEL assay to calculate the apoptotic index(AI).Result Compared with the control group,all the poisoned rats showed evident pathological damage to the heart muscles and the AI significantly increased in Atropine group and Fasudil group(P0.01).Compared with Atropine group,Fasudil group showed a higher survival rate,attenuated pathological damage to the heart muscles and significantly decreased AI(P0.01).Conclusion Fasudil could elevate the survival rate of the rats with acute omethoate poisoning,markedly reduced the myocardial damages and significantly inhibited the myocardiocyte apoptosis.Therefore,fasudil could provide protective effect on the myocardial damage in rats with acute omethoate poisoning.
