嘌呤核苷酸对海洛因处理过大鼠C6神经胶质瘤细胞增殖的影响

摘要 目的探讨海洛因及嘌呤核苷酸对大鼠C6神经胶质瘤细胞增殖的影响。方法体外培养大鼠C6神经胶质瘤细胞,分别以不同浓度的海洛因及嘌呤核苷酸处理细胞,MTT比色法检测海洛因和嘌呤核苷酸对细胞增殖的影响。结果 MTT实验表明,海洛因对大鼠C6神经胶质瘤细胞的增殖有抑制作用,120mg/L海洛因作用24h对C6细胞的抑制率达52%,并表现为剂量依赖性;嘌呤核苷酸对C6细胞的增殖有促进作用,120mg/L嘌呤核苷酸作用24h,细胞增殖率为30%,并表现为剂量依赖性。结论海洛因抑制大鼠C6神经胶质瘤细胞的增殖;嘌呤核苷酸促进大鼠C6神经胶质瘤细胞的增殖,并在某种程度上对抗海洛因对C6细胞增殖的抑制作用。 Objective To investigate the effects of heroin and purine nucleotides on the proliferation of C6 glioma cells. Methods The effects of heroin and purine nucleotides on the proliferation of C6 glioma cells were evaluated by MTT assay. Results Heroin inhibited the growth of C6 glioma cells in vitro,52% C6 glioma cells were inhibited when the ceils were treated with 120 mg/L heroin for 24 h;purine nucleotides promoted the proliferation of C6 glioma cells,30% C6 glioma cells were promoted when the cells were treated with 120 mg/L purine nucleotides for 24 h. Conclusions Heroin inhibits the proliferation of C6 glioma cells, purine nucleotides promotes the proliferation of C6 glioma cells.
