相较正常结肠黏膜,Hath1在非粘液性结肠腺癌中的表达下调 被引量:4

摘要 目的:研究Hath1基因在非粘液性结肠癌中的表达。方法:从48例非粘液性结肠腺癌患者的标本中取材,石蜡包埋切片,H&E染色,观察正常结肠黏膜、癌旁黏膜和结肠癌组织中杯状细胞数量的变化。用定量实时RT-PCR和Western blot的方法,检测这3种组织中Hath1蛋白和Muc2蛋白的表达。结果:在结肠癌组织中未发现杯状细胞;结肠癌组织中Hath1 mRNA、Hath1蛋白、Muc2 mRNA和Muc2蛋白的表达均显著下调。结论:Hath1基因的表达下调,可能与非粘液性结肠腺癌的发生有关。 Objective:To investigate the Hath1 expression in colon non-mucinous adenocarcinomas.Methods:Sections of paraffinembedded tissues were used to investigate the goblet cell population of normal colon mucosa,mucosa adjacent colon cancer and colon cancer samples which came from 48 patients with colon non-mucinous adenocarcinoma.Hath1 and Muc2 expression in these samples were tested by immunohistochemistry,quantitative real-time reverse transcription-PCR and Western blot.Results:There were not any goblet cell to be found in colon cancr.The levels of Hath1 mRNA and Hath1,Muc2 mRNA and Muc2 were significantly down-regulated in colon cancer.Conclusion:The down-regulation of Hath1 expression may correlate with colon carcinogenesis.
