摘要
目的:研究不同强度脉冲电磁场(PEMFs)对大鼠成骨细胞(ROB)增殖与分化的影响。方法:酶消化法分离获得大鼠颅骨成骨细胞,采用50 Hz,0.14、0.16和0.18 mT的PEMFs进行干预,测定细胞增殖率、ALP活性和BMP-2 mRNA的表达。结果:0.14 mT的PEMFs对ROB增殖和分化均无明显影响;0.16、0.18 mT的PEMFs可明显提高ALP活性,并上调BMP-2mRNA表达。结论:PEMFs对ROB的影响主要表现在促进其分化,且这种作用存在较为敏感的"强度窗"效应。
Objective:To investigate the effects of different-intensity pulsed electromagnetic fields(PEMFs) on the proliferation and differentiation of rat osteoblasts(ROB).Methods:The rat calvaria osteoblasts were separated with enzymatic digestion method,stimulated by the PEMFs at 50 Hz with 0.14 mT,0.16 mT or 0.18 mT.The proliferation rate of osteoblasts,ALP activity and BMP-2 mRNA expression were determined.Results:The PEMFs at 50 Hz with 0.14 mT had no appreciable effect on the proliferation and differentiation of ROB.The PEMFs at 50 Hz with 0.16 mT and 0.18 mT significantly increased ALP activity and BMP-2 mRNA expression.Conclusion:The PEMFs can significantly stimulate differentiation of ROB,with sensitive "windows" effects.
