肾上腺皮质腺瘤及嗜铬细胞瘤组织中NF-κB与iNOS蛋白表达水平研究

摘要 目的探讨核转录因子κB与诱导型一氧化氮合酶在人肾上腺皮质腺瘤及嗜铬细胞瘤组织中的表达差异。方法采用免疫组织化学两步法检测13例肾上腺皮质腺瘤及15例嗜铬细胞瘤组织标本中NF-κB与iNOS蛋白的表达,以染色后阳性细胞百分比来评价表达强度,并进行统计学分析。结果人肾上腺皮质腺瘤及嗜铬细胞瘤组织中NF-κB与iNOS蛋白水平明显高于正常组(P均<0.05);嗜铬细胞瘤组中NF-κB与iNOS蛋白水平明显高于肾上腺皮质腺瘤组(P<0.05),各组中NF-κB与iNOS蛋白之间的表达呈正相关(P<0.05)。结论人肾上腺皮质腺瘤和嗜铬细胞瘤组织中,高表达的NF-κB和iNOS与其发生发展密切相关;在肾上腺皮质腺瘤及嗜铬细胞瘤组织间,NF-κB与iNOS蛋白表达有显著差异,提示肾上腺腺瘤和嗜铬细胞瘤在发病机制中可能有着本质差别;而NF-κB的活化与iNOS的表达,在发病过程中相互作用、相互制约。 Objective To investigate the expressions of nuclear transcription facror κB(NF-κB)and inducible nitric oxide synthase(iNOS)in adrenocortical adenoma and pheochromocytoma tissue.Methods The expression of NF-κB and iNOS protein was detected in 13 cases of adrenal cortical adenoma and 15 cases of pheochromocytoma tissues by immunohistochemistry(two-step method)in order that the expression intensity was evaluated by the percentage of staining positive cells.Results The expressions of NF-κB and iNOS protein in adrenocortical adenoma and pheochromocytoma tissue were higher than that in nomal tissue(P〈0.05);the expressions of NF-κB and iNOS protein in pheochromocytoma tissue were significantly higher than that in normal group(P〈0.05);the experssion of NF-κB was positively correlated with that of iNOS(P〈0.05).Conclusion The overexpression of NF-κB and iNOS-mRNA plays an important role in the development of adrenocortical adenoma and pheochromocytoma.The significant difference in NF-κB and iNOS protein between adrenocortical adenoma and pheochromocytoma indicated that they may be different in nature in the pathogenesis.The expression of iNOS and activation of NF-κB influence and restrict each other.
