摘要
本研究以优良品系翠绿1号假俭草的种子为外植体,成功地建立了再生体系。基本培养基为MS,在添加了2,4-D 1.5 mg/L的培养基上最适于种子愈伤诱导,愈伤诱导率为99%,黄色颗粒愈伤诱导率为4.8%,愈伤最佳继代培养基为MS+2,4-D 1.5 mg/L;在最佳分化培养基MS+KT 2.5 mg/L上,黄色愈伤分化率为99%,芽最佳生长培养基为MS+BA2.0 mg/L+NAA0.7 mg/L;试管苗最佳生根培养基为MS+NAA0.6 mg/L,生根率达到99%;生根的试管苗移栽到装有田园土的土盆中,其存活率达到88%。
Plant regeneration systems have been successfully established by using the seed of centipedegrass Cuilv No.1' as explant.The basal culture medium was MS,the optimum medium of callus induction was MS medium with 2,4-D1.5 mg/L,the rate of callus induction was 99%,The rate of yellow granule callus induction could be as high as 4.8%,the optimum medium of callus subculture was MS medium with2,4-D 1.5 mg/L;The rate of yellow granule callus differentiation could be as high as 99% on MS medium plus KT 2.5 mg/L,the optimum medium of shoot growth was MS medium plus BAP 2.0 mg/L and NAA 0.7 mg/L;and The rooting rate was 98% on MS rooting medium pus KT 2.5 mg/L.The survival rate of plant transplanting was 92% after transferred to pots with garden soil.
