摘要 目的探讨抑癌基因PTEN(第10号染色体缺失的磷酸酶和张力蛋白同源基因)在胃癌组织中的表达与患者预后的关系。方法采用免疫组织化学SABC法比较92例胃癌组织和35例正常胃黏膜中PTEN表达差异,随访胃癌术后病人的生存率。结果正常胃黏膜中,PTEN全部强阳性表达。胃癌组织中,PTEN蛋白的表达和着色呈异质性,高分化癌尚有部分表达,低分化癌或转移癌表达明显下降,甚至不表达。随访PTEN阳性表达组生存率高于PTEN蛋白阴性表达组。结论 PTEN在胃癌中的表达降低或缺失,可能成为预测胃癌病人预后的一种有价值的分子标记物。 Objective To explore the relationship of PTEN protein expression and prognosis in gastric carcinoma. Methods The expression of PTEN protein in 92 cases of gastric carcinoma and 35 cases of normal gastric mucosa was detected by immunohistochemical SABC method.The patients were followed up after surgery and the survival rate was analyzed. Results The strong positive expression was found in all normal gastric mucosa,while the heterogeneity expression in gastric carcinoma.The PTEN protein partly expressed in well-differentiated gastric mucosa,while the PTEN protein decreased to express or negatively expressed in poorly differentiated carcinoma and metastatic carcinoma.The survival rate of positive expression patients was higher than that of negative expression patients. Conclusion PTEN protein expression decrease or deficiency could be used as an important molecular marker for predicting of gastric carcinoma.
