摘要
目的:研究比较莪术油、莪术残油(莪术油提取出β-榄香烯的残余物)在大鼠血浆中莪术醇和莪术二酮的含量。方法:采取大鼠灌胃给药,以对羟基苯甲酸乙酯为内标物,采用毛细管气相色谱法同时测定,内标法计算莪术油、莪术残油中标志性成分莪术醇和莪术二酮的血浆药物浓度,运用DAS2.0分析计算药代动力学参数。结果:莪术醇和莪术二酮分别在0.001028~0.6580mg/mL和0.04064~1.300mg/mL范围内呈现良好的线性关系,精密度、稳定性及融冻次数考察均符合有关要求,测定方法的绝对回收率莪术醇为72.3%~85.3%,莪术二酮为70.1%~87.0%;内标物对羟基苯甲酸乙酯的绝对回收率为74.9%~96.5%;方法回收率莪术醇为55.8%~69.0%,莪术二酮为61.6%~68.3%,均符合有关要求。通过药时曲线和药代动力学参数分析,莪术油、莪术残油中莪术醇、莪术二酮在大鼠的体内代谢过程基本符合二房室模型。结论:在莪术油和莪术残油中莪术醇的血药浓度变化相似,而莪术二酮血药浓度存在较大差异。
AIM: To compare the content difference in rat's plasma concentration of curcumol and curdione between zedoary oil and its residue oil(the latter come from residum after removing β-elemene.METHODS: After the zedoary oil and its residue oil was administered,ethyl-p-hydroxybenzoate in rat's plasma used as internal standard was carried out simultaneously to determine curcumol and curdione by capillary column gas chronatography.RESULTS: A good linearity was obtained when the concentration of Curcumol ranged from 0.001 028 to 0.658 0 mg/mL,and the concentration of Curdione ranged from 0.040 64 mg/mL to 1.300 mg/mL.Precision,stability and the study of the number of thawing and freezing test were within the requirements.The recovery of curcumol was 72.3%-85.3% ,the recovery of Curdione was 70.1%-87% .The recovery of internal indicator was in the range of 74.9%-96.5% .The Curcumol’s methods recovery was in the range of 55.8%-69% ,the Curdione’s methods recovery was in the range of 61.6%-68.3% .Recovery experiments were within the requirement.Two-compartment model of curcumol and curdione was fit to metabolic processes in vivo in rats drug concentration-time curve and pharmacokinetic parameters.CONCLUSION: Curcumol’s changes of both zedoary oil and residue oil in plasma concentration are similar in blood concentration,but curdione’s changes shows great difference.Two indicators make difference in pharmacokinetic parameters.
