摘要
采用高效液相色谱技术,分离制备高纯度银杏内酯。通过单因素及正交试验,对银杏内酯的浸提条件进行优化,再通过反相高效液相色谱分离系统,采用浓度梯度洗脱的方式,分离出高纯度银杏内酯。结果显示最佳浸提条件:提取剂为乙酸乙酯,浸提时间4h,温度65℃,固液比1:5,银杏内酯含量由原料中的6%,提高到29%。通过制备色谱柱层析,得到产品中银杏内酯纯度≥90%,回收率为70%。
Separation and preparation of high purity of ginkgolides from Ginkgo biloba leaves was investigated by high-performance liquid chromatography (HPLC). The efficient factors of extraction of ginkgolides by single factor tests and orthogonal experiment were optimized. RP-HPLC using gradient elution method was utilized. And then the high purity ginkgolides was achieved from the crude extract of Ginkgolide. Results showed: the optimum conditions of extraction as follows: the extractant of ethyl acetate, extraction temperature 65~(3, extraction time 4 h, ratio of solid to liquid extraction 1:5. The content of ginkgolides was increased from 6% to 29%. The product of ginkgolides was obtained by RP-HPLC, product purity was about 90%, product yield was 70%.
