摘要
目的探索牛奶中残留药物的筛查方法。方法建立牛奶的HPLC-MS指纹图谱,经XCMS软件处理,提取特征信息(特征离子和对应的保留时间);以50份原料奶为样本,以特征信息为参数建立牛奶成分的本底数据库;从样品分析后的特征信息中扣除本底数据库中已有的参数值,对异常参数值进行分析、确证,找出异常化合物。结果通过标准添加实验,筛查出添加的有害药物,验证了筛查方法的可行性;通过对市售牛奶样品的筛查,发现了异常化合物,经分析为性激素雌二醇或其衍生物。结论本文建立的方法是一种较全面、可行、应用性广的牛奶中残留药物筛查技术。
Objective To screen hazardous drug residues in milk.Methods HPLC-MS fingerprints of milk were established and then processed by XCMS software to acquire characteristic information,including characteristic ion andcorresponding retentionti me.The background database about milkingredients was developed by taking 50 raw materialmilk as the speci mens and characteristic information as parameters.When the milk sample was analysed,the charac-teristic information which was already in the background database was deducted fromthe parameter database of milksample,and surplus characteristic information was considered abnormal parameters and then analysed and validated toscreen abnormal compounds,which might be hazardous drug or a detri ment.The spiked experi ment was carried out tovalidate the feasibility of screening method,and the method was applied to screen detri ments in commercially availablemilk samples.Results Spiked hazardous drugs were screened out,indicating that the method was feasible.An ab-normal compound was also screened out fromthe commercial pure milk with the method.After analysis andidentifica-tion,the abnormal compound was the sex hormone estradiol.Conclusion The method is global,feasible,practical,and can be applied to screen detri ments in commercially available milk samples.
