虾头蛋白制降血压肽酶解条件及分子量分布研究 被引量:3

Study on hydrolysis condition and molecular weight distribution of antihypertensive peptide derived from head protein

摘要 利用碱性蛋白酶酶解南美白虾虾头蛋白制备具有血管紧张素转移酶(ACE)抑制活性的降血压肽,进而采用凝胶过滤色谱法探讨酶解产物的分子量分布。结果表明:在50℃、pH8.5、酶量0.3%(w/w)、酶解2 h条件下,所得酶解产物的ACE抑制率最高。此条件下酶解产物经SephadexG-15凝胶过滤,色谱图上出现5个吸收峰,其中高ACE抑制活性组分的分子量分布在262~470之间,IC50为0.79 mg/mL。 Alcalase Protease was employed to hydrolyze Penaeus Vannamei head protein to prepare peptide with angiotensin Ⅰ-converting enzyme inhibitory activity.Gel column filtration chromatography was used to determine the molecular distribution of the hydrolysate.The results showed that the hydrolysate with the highest inhibition ratio was prepared under the condition of 50 ℃,pH8.5,enzyme concentration 0.3%(w/w) and 2 h.Gel filtration of the hydrolysate on Sephadex G-15 chromatogram yielded five absorbance peaks,the molecular distribution of the fractions with the most potent inhibition activity was located between 262~470,the IC50 was 0.79 mg/mL.
