摘要
目的观察牛磺酸(Tau)对染铅大鼠肝细胞膜ATP酶活力及肝功能的影响。方法把50只Wistar大鼠随机分为正常对照组、染铅组、Tau低剂量治疗组、Tau中剂量治疗组、Tau高剂量治疗组,每组10只动物。对照组用蒸馏水灌胃,其余组用6g/L醋酸铅溶液按每天60mg/kg灌胃;染毒30d后,Tau低、中、高治疗组动物分别用200、400、800mg/kg的Tau每天灌胃1次,正常对照组及染铅组用等容量蒸馏水灌胃,连续30d后,测定血铅水平、血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)活力和肝细胞膜Na+-K+ATP酶和Ca2+-Mg2+ATP酶活力。结果 Tau各剂量组(200、400、800mg·kg-1·d-1)大鼠肝细胞膜Na+-K+ATP酶和Ca2+-Mg2+ATP酶活力明显升高(P<0.05或P<0.01)。而对铅所致大鼠血清ALT和AST的明显升高均有不同的降低作用(P<0.05或P<0.01)。结论 Tau对铅染毒大鼠肝细胞ATP酶活力及肝功能的损伤具有明显的保护作用。
Objective To observe the effect of taurine on membrane ATPase activities of hepatocytes and liver function in rats exposed to lead.Methods 50 rats were randomly divided into 5 groups:negative group,positive group and therapeutic group of taurine at the doses of 200,400 and 800 mg·kg-1·d-1.The negative group was gavaged with distilled water while other groups with 6 g/L lead acetate solution at 60 mg·kg-1·d-1 for 30 days.Then the therapeutic groups were gavaged with taurine solution at 200,400,800 mg·kg-1·d-1 and other groups with distilled water for 30 days.Lead contents of blood were measured by atomic absorption spectrometry (AAS),ALT and AST were measured by automatic chemistry analyzer (ACA) and membrane ATPase activities of hepatocytes were measured by a reagent kit.Results The activities of Na+-K+ATPase and Ca2+-Mg2+ATPase in all dose groups were obviously higher than those of the positive group (P0.05,P0.01).The raise of serum ALT and AST was significantly depressed in all dose groups (P0.05,P0.01).Conclusion Taurine could prevent membrane ATPase of hepatocytes from lead injury.
