摘要
目的:观察胰岛素对局灶缺血脑组织cfos基因表达的影响。方法:30只肾血管性高血压大鼠(RHR)随机分4组,在大脑中动脉闭塞(MCAO)后即予胰岛素(2.1IU/kg,A组)、葡萄糖(2g/kg)和低剂量胰岛素(2.1IU/kg,B组)、葡萄糖(4g/kg)和高剂量胰岛素(4.5IU/kg,C组)、生理盐水(D组),采用原位杂交技术检测各组MCAO后3h脑组织cfos基因的表达。结果:低剂量胰岛素组(A、B组)cfosmRNA未出现明显变化(P>005),而C组cfosmRNA则有统计学意义的增加(P<001)。结论:胰岛素促进缺血脑组织cfos基因表达,可能是其神经保护作用机制之一。
Objective: To investigate the effect of insulin on c fos gene expression in focal ischemic rat brain. Methods: The renovascular hypertensive rats(RHR) (n=30) were divided at random into 4 groups which received separately with insulin only (group A, 2.1IU/kg),insulin and glucose (group B with low dose insulin, 2.1IU/kg; group C with high dose insulin, 4.5IU/kg) or normal saline (group D) immediately after middle cerebral artery occlusion (MCAO). Level of c fos mRNA on brain sections was measured by in situ hybridization (ISH) at 3h after MCAO. Results:Little effect on c fos expression was detected in groups with low dose of insulin (group A, B) (P>0.05) and a significant increase was detected in group C(P<0.01). Conclusions: These results indicate that the effect of insulin on c fos expression may be one of the mechanisms of neuroprotection of insulin.
