摘要
为了探讨血吸虫重组BCG-Sj26GST疫苗免疫鼠后对尾蚴攻击感染的保护性作用,采用106CFU和108CFU重组BCG-Sj26GST疫苗皮下接种BALB/C鼠,接种后8周用日本血吸虫尾蚴进行攻击感染,感染后6周剖杀小鼠,计算减虫率、减卵率,同时进行虫卵肉芽肿周长和面积测量,以及抗体水平和抗原水平测定,同时设有PBS对照组。结果表明重组BCG-Sj26GST疫苗免疫小鼠后,106CFU、108CFU组与对照组相比,减虫率分别为27.34%、16.64%,减卵率分别为55.75%、60.50%,虫卵肉芽肿的平均周长和平均面积、免疫后血清抗体水平差别均具有非常显著意义;108CFU与106CFU组相比,血清抗原水平差别具有显著意义,说明血吸虫重组BCG-Sj26GST疫苗是一种比较理想的新型疫苗。
In order to investigate the protection against challenge with cercarie of schistosoma japonicum, by immunization with recombinant BCG Sj26GST vaccine, BALB/C mice were immunized subcutaneously by 10 6CFU and 10 8CFU rBCG Sj26GST vaccine of schistosome respectively, challenged with Sj cercarie on the 8th week of inoculation and killed on the 6th week of infection to count rate of reduced worm burdens, rate of reduced eggs number, to measure the circumference and the area of egg granulomas,and to determine the level of antibody and circulating antigen. At the same time, PBS served as control group. The results showed that after the mice were immunized with the vaccines, the rate of reduced worm burdens and the rate of reduced eggs number were 27 36 % vs 16 64 % and 55 75 % vs 60 50 % respectively in the experimental group and control groups, and average circumference, average area and the level of antibody had a significant difference between the groups. The level of circulating antigen showed a marked difference between 10 8 CFU and 10 6 CFU groups. It is suggested that rBCG Sj26GST vaccine of schistosome is a new type of more ideal vaccine.
