摘要
使用束缚方法使大鼠产生应激,一部分实验取大鼠应激3h、6h、12h和18h的淋巴结提取物,与刀豆素A(ConA)同时加入到正常大鼠肠系膜淋巴结细胞悬液中,浓度比为1∶160和1∶640。另一部分实验取大鼠应激3h、6h、12h、18h和解除束缚后12h、24h、48h、72h、96h的血清,与ConA同时加入到正常大鼠肠系膜淋巴结细胞悬液中,浓度比为1∶16和1∶64,孵育68h后用噻唑蓝(MTT)比色分析法检测肠系膜淋巴结T细胞增殖的光密度(OD)值。旨在说明应激抑制T细胞增殖的出现、加强和恢复过程。结果如下:(1)应激3h和6h大鼠的淋巴结提取物和血清对T细胞的增殖都没有明显的影响。(2)应激12h,两种浓度的淋巴结提取物和血清都出现对T细胞增殖的明显抑制作用,应激18h的作用更强。(3)束缚解除后,血清对淋巴细胞的抑制作用仍然存在,但随着时间的延长抑制作用逐渐减弱,至束缚解除后96h,抑制作用基本消失。上述结果提示,大鼠束缚应激能明显抑制T淋巴细胞的增殖;这种抑制作用需要束缚应激一段时间后才能出现;并且在解除束缚后仍能持续较长时间。
In the present study,
restraint, which causes no direct tissue damage, was selected as a stressor to investigate the
suppressive effect of the restraint stress on the T-lymphocyte proliferation in rats. The extract
from the lymph node, which was respectively drawn at 3 hour, 6 hour, 12 hour, 18 hour of
restraint stress and the serum, which was respectively drawn at 3 hour, 6 hour, 12 hour, 18
hour of restraint stress and at 12 hour, 24 hour, 48 hour, 72 hour, 96 hour after the stress, were
added to the lymphocyte suspension of mesenteric lymphaden of rat with concanavalin A (Con
A) and were incubated for 68 hours. The optical density (OD) values, which show the level of
T-lymphocyte proliferation, were determined by means of the colorimetric assay of
methylthiazol tetrazolium bromide (MTT). The results are as follows:(1)The extract from the
lymph node and the serum drawn at 3h and 6h of stress did not markedly affect the
T-lymphocyte proliferation.(2)At 12h of stress, the extract from the lymph node and the serum
significantly inhibited the lymphocyte transformation, the inhibition being stronger at 18h of
stress.(3)After the restraint stress, the suppressive effect of serum on the T-lymphocyte
proliferation gradually decreased, and to 96h after the stress, the suppression basically
disappeared.The above results suggest that restraint stress may cause the inhibition of
T-lymphocyte proliferation, and it takes some time for the inhibitory effect to appear and tonger
time to disappear.
