摘要
目的利用流式细胞术多参数分析的特点观察慢性阻塞性肺病(COPD)大鼠肺组织局部树突状细胞(DC)的含量。方法通过烟熏法3个月建立慢性阻塞性肺气肿大鼠模型;于第4个月处死,一方面肺组织切片HE染色观察病理改变;另一方面将肺组织用机械法制备成单个细胞悬液,调整细胞浓度为5×106个细胞/ml。在模型组、正常对照组中均加入CD11c、CD80两种抗体,然后用流式细胞仪分别检测两组双色标记的阳性百分率。结果 COPD模型组肺组织中出现COPD的特征性病理改变;且COPD组中CD11c及CD80双阳性率为(1.87±0.23)%,明显高于空白对照组(0.56±0.12)%,两组差异有统计学意义(P<0.01)。结论 COPD动物模型组中DC数量明显升高,提示DC在COPD中发挥着重要作用。
Objective To count and analyze accurately the dendritic cells(DC)of lung tissue in rats with chronic obstructive pulmonary disease(COPD)by flow cytometry.Methods Thirty male Wistar rats were randomly divided into two groups(n=15):control group and COPD group.The COPD rat models were established by fumigation(3 months).The lung tissues were taken to pathological examination by HE staining,and were simultaneously prepared by mechanical means into a single cell suspension.Adjusted the cell concentration to 5×10^6 cells/ml.Every group was added both anti-rat CD11c and anti-rat CD80,which were monitored by flow cytometry.Results The COPD model group had the pathological changes of COPD,and its double-positive percentage of CD11c and CD83 expression was(1.87±0.23)%,which was dramatically higher than that of control group(0.56±0.12)%.Statistical analysis showed there was significant difference between two groups(P〈0.01).Conclusion The number of DC in the COPD model group is significantly higher than that in the control group,and implies that the pulmonary DCs play an important role in the pathogenesis of COPD.
