摘要
目的通过研究细菌毒素对人腹膜间皮细胞受体整合素分布及细胞骨架的影响,探讨炎症引起间皮细胞脱落的机制。方法分离人腹膜间皮细胞作体外培养,采用活细胞荧光抗体染色,共聚焦显微镜断层扫描的方法,检测细胞整合素α3β1的分布;以考马斯亮兰染色法,观察细胞骨架结构的变化。结果正常培养的人腹膜间皮细胞表达受体整合素α3β1,其分布由细胞底层至顶层逐渐减少;而脂多糖(LPS)或肠毒素刺激20分钟后,测得整合素α3β1的分布由底部至顶端转移。结论细菌毒素引起的人腹膜间皮细胞受体整合素α3β1分布和细胞骨架改变可能是引起间皮细胞脱落的原因之一。
Objective To study the effects of bacterial toxin on the
redistribution of integrin31 and cytoskeleton on the human peritoneal mesothelial cell(HPMC).
Methods Mesothelial cells from human omenta were isolated and cultured in vitro.The second
passages were seeded in Petri dishes.The distribution of integrin31 on HPMC was detected by
indirect immunofluorescence using confocal microscope and the structure of cytoskeleton was
observed by Comas Blue stain. ResultsBZNormal human HPMC expressed integrin31 and the
maximal intensity of fluoresence was focused on basal layer of the cells.After stimulated with
LPS or enterotoxin for 20 minutes,the average fluorescence levels decreased in basal,but
increased in apical layers.The ratio of fluorescence intensity between apical and basal layers
was significantly increased in HPMC stimulated with bacterial toxins compared with those
incubated with medium alone. Conclusion The redistribution of receptor integrin31 and the
altered cytoskeleton induced by bacterial toxin is responsible for the detachment of HPMC in
bacterial peritonitis.
