摘要
目的探讨钒酸钠改善创伤后胰岛素抵抗和糖代谢紊乱的作用机制。方法用钒酸钠胃饲30%TBSAⅢ度烫伤大鼠,观察血糖、胰岛素、乳酸动态变化,糖原储存和脂肪合成的变化,以及对胰岛素受体酪氨酸蛋白激酶(IR-TPK)活性的影响。结果钒酸钠降低严重烫伤大鼠高血糖、高胰岛素和高乳酸血症,增加肌肉和肝脏糖原储存及胰岛素刺激的脂肪合成,对骨骼肌细胞膜胰岛素受体影响不明显,但可明显恢复IR-TPK活性。结论钒酸钠可明显改善严重烫伤大鼠胰岛素抵抗及糖代谢紊乱,其机制不涉及胰岛素受体,而是直接刺激肌肉细胞膜IR-TPK活性。
Objective To Clarify the mechanism of vanadate stroving insulin resistanceand glucose metabolism disorder in severely scalded rats. Methods After vanadate adlninistrated the ratS with 30% TBSA scald, the chance of Plasma index, glycogrn content andlipognesis were oberved. The changes of insulin receptor binding aCtivity and its tyrosineprotein kinase activity were also observed. Results Vanadat6 could decrease hyperglycemiaand hyperlactacidelnia, increase the contact of muscle glycogn and liver giycogrn, and theamount of lipoensis by insulin speculation. The receptor itself was not Chance. ConclusionVanadat6 can Significantly approve the insulin resistance and glucose metabolism disorder, butthe mechanismal has no relation with insulin reCeptor, it Stimulates directly the TPK activityof myocyte membrane.
