同种异体皮质骨修复过程中胶原基因的表达及其应力适应性 被引量:5

摘要 目的探讨同种异体皮质骨修复过程中胶原基因的表达情况及其应力适应性。方法制造兔双侧尺骨缺损1.0cm,并行骨移植。后一侧承受正常生理载荷,另侧承受低载荷的动物模型;利用核酸分子原位杂交技术,分阶段比较同种异体皮质骨修复过程中Ⅰ、Ⅱ、Ⅲ型胶原基因的表达情况。结果同种异体皮质骨植入后,植入骨一宿主骨界面间连接的方式是在纤维骨痴的基础上直接成骨。愈合过程中只有Ⅰ、Ⅲ型胶原基因的表达。移植骨内部修复是先出现大量吸收腔,继而在吸收腔内出现大量表达1型胶原基因的细胞。从第4周开始,在移植骨内部的骨吸收陷窝内,正常载荷侧1型胶原的基因表达比低载荷侧更为明显。结论不同载荷可以影响移植骨修复过程中胶原基因的表达方式。 Objective To explore the collage gene expression Under differal biomechanical environments in the incompration pram of conical ailogrsft. Methods A rabbitmodel of our own design with tWo transplanted alloarefts was subjeded to differentloading Patterns. Specimens were taken for analysis of collage gene opndon by in situcDNA-mRNA hybridhation at different stsgn. Results The union Pattern of anografthost bone junCtion was fibrous callus formation followed by direct bone formation withtype Ⅰ and type Ⅲ collage gene opedon. The remodeiling of graft Proceeded initiallymarked with osteoclast resorption followed by the appearing of preostedblast and osteoblast which expressed my Ⅰ collage gene in the resoghve cavity. In comparison ofnormally loaded grsft with that of low loaded daft, there was more ostedblast-like cellswith the opredon of for I collage gene occurring in the nit6rface at early Stage andinterior at later Stabs SinCe the 4th week. Conclusion The mechanichl load can regulate thepattern of collage gene opredon.
