bFGF mRNA的转录与喉粘膜良性及恶性病变的关系 被引量:3

Relationship between the mRNA transcription of basic fibroblast growth factor (bFGF) and benign and malignant laryngeal mucosa

摘要 目的:探讨碱性成纤维细胞生长因子(bFG)与喉癌生长、浸润、转移的关系及其可能的作用机制.方法:用原位杂交的方法观察bFGFmRNA在正常喉粘膜上皮(N)、慢性非特异性炎症(IF)、不典型增生(DYS)及鳞癌(SCC)中的表达.结果:在N,IF.DYS及SCC的间质中均检测到bFGFmRNA的表达.上皮细胞未见阳性表达.在N及IF,阳性反应部位位于血管内皮细胞、成纤维细胞;IF的阳性反应略有提高,此外部分炎细胞也见阳性反应;DYS及SCC中的阳性反应程度较N及IF有所增强,尤其是新生血管内皮细胞的阳性率及反应程度明显提高.在SCC中,分化差的bFGF表达较分化好的SCC阳性反应程度强.结论:bFGF不仅可以促进喉癌新生血管及间质的形成.而且可能参与肿瘤细胞生长.在喉癌中,bFGF以旁分泌生长方式影响肿瘤的生长. AIM: To determine the distribution and functionof bFGF in normal and benign condition and the carcinogenesis of laryngeal mucosa. METHODS: the localization andtranscriptional level of bFGF mRNA in normal (N ). inflammation (IF ). dysplasia (DYS ) and squamous cell carcinoma(SCC ) were assessed by in situ hybridization. RESULTS:bFGF mRNA was expressed by the mesenchymal cells in N.IF. DYS and SCC groups. whereas. there was not any posi-tive reaction in epithelial cells. fibroblasts and endothelialcells and inflammatory cells. but not by any of the epithelialcells. The expression of bFGF was defected in endothelialcells, fibroblasts and chronic inflammatory cells. There is anincreasing tendency of mRNA transcriptional level from N,IF, DYS to SCC. In SCC, the newly formed blood vesselswas particularly intensive labeled. Furthermore, the expression level was higher in poorly differentiated SCC than welldifferentiated SCC. CONCLUSION: bFGF was highly associated with the neovascularisation and formation of stroma. hplays an important role in the epithelial--mesenchymal interaction. bFGF might be one of the critical factors for maintainthe structure and metabolism of normal mucosa and thegrowth of laryngeal carcinoma.
