摘要
参照《中国药典》2000年版一部附录XD挥发油测定法提取马尾松松针挥发油;利用乌头碱诱发大鼠心律失常法,建立药物动力学模型;测定松针挥发油对大鼠心肌外膜单相动作电位的单相动作电位幅度、去极化时程、动作电位最大上升速率、50%复极化动作电位时程和90%复极化动作电位时程的影响。结果松针挥发油中(0.226 mL/kg)、高剂量(0.715 mL/kg)能显著对抗乌头碱致大鼠心肌单相动作电位APD90的缩短作用(P<0.05),低(0.072 mL/kg)、高(0.715 mL/kg)剂量能显著对抗乌头碱致Vm ax的上升作用(与阴性组比P<0.05)。结果证明马尾松松针挥发油对药物诱发心律失常具对抗作用,此作用是通过影响心肌细胞的离子分布而实现;松针挥发油能阻止乌头碱所致的钠离子内流作用。
Pine needle oil was distilled from Pinus massoniana Lamb.with a reference to Appendix XD of Chinese Pharmacopoeia(edition 2000).Aconitine was used to establish rat arrhythmia model,then the effects of pine needle oil on monophasic action potential amplitude,duration of depolarization,maximum velocity of 0 phase depolarization(Vmax),action potential duration at 50% and action potential duration at 90%(APD90) was investigated by measuring monophasic action potential amplitude of outward membrance of rats heart,meanwhile it's action mechanism on arrhythmia was studied.The results showed that pine needle oil from Pinus massoniana Lamb.at 0.226 mL/kg and 0.715 mL/kg could significantly inhibit Aconitine-induced APD90 shortening(P0.05),0.072 mL/kg and 0.715 mL/kg could significantly inhibit Aconitine-induced Vmax increasing(P0.05),compared with the negative group).It is concluded that pine needle oil from Pinus massoniana Lamb.could resist the arrhythmias induced by some drugs through its effects on ions' distribution in the heart.It could also inhibit inward sodium current which was induced by Aconitine.
