摘要 肿瘤的发生和发展是一种多基因参与,多因素影响的多步骤复杂过程。主要表现为肿瘤细胞增殖失控,分化障碍及其凋亡受阻。生理情况下,细胞的增殖、分化、成熟及凋亡受到多种正负调控因子的严格的调控,正、负调节信号处于动态平衡中,维持着造血的稳定。肿瘤发生的机制之一可能是细胞对负反馈抑制物的反应性发生改变而使细胞逃脱了正常生长调控。近年来的许多研究表明TGF-β信号传导通路的异常与许多肿瘤的发生、发展密切相关,TGF-β1、TGF-β-RⅠ、TGF-βR-Ⅱ、Smads、靶基因的质和量的改变都和人类肿瘤密切相关。现对TGF-β信号转导通路受体、因子及下游基因与肿瘤的关系做概括。 The occurrence and development of tumor is a multi - gene involvement, many factors affect the multi - step complex process. Mainly the control of tumor cell proliferation,differentiation and apoptosis was blocked. In physiological conditions,coil proliferation, differentiation, maturation and apoptusis was controlled by a variety of positive and negative regulators, they keep in a dynamic balance, maintaining the stability of blood environment. One mechanism of leukemia pathogenesis was negative feedback inhibitor of reactive cells occurs change it to escape normal growth control. Recently, many studies show that TGF - β signaling pathway abnormalities was closely related to the occurrence and development of tumors,TGF - β1 ,TGF - β - R I ,TGF - βR - II ,Smads,turget gene changes in the quality and quantity were also closely related to human tumor.
