摘要
目的:建立离子对高效液相色谱法检测中成药及保健食品中添加二甲双胍、苯乙双胍、格列齐特、格列美脲、格列本脲、格列吡嗪、甲苯磺丁脲、罗格列酮、吡格列酮共9种化学降糖药。方法:采用AgilentZORBAX-EclipseXDB-18(4.6mm×250mm,5μm)色谱柱分离,流动相为0.02mol·L-1磷酸二氢钾溶液(含2mmol.L-1十二烷基硫酸钠)-乙腈(62.5∶37.5)(用1mol.L-1氢氧化钠溶液调节pH至6.5),流速1.0mL·min-1,检测波长230nm,柱温30℃。结果:9种化学降糖药能完全分离;具有宽的线性范围和良好的线性关系(r>0.9992),检出限为5.9~43.5μg.L-1。结论:该方法用于中药及保健食品中化学降糖药的检测具有快速、简便、高效的特点。
Objective:To develop an ion-pair HPLC method for the simultaneous estimation of nine anti-diabetic agent——metformin,phenformin,gliclazide,glimepiride,glibenclamide,glipizide,tolbutamide,rosiglitazone,pioglitazone,which is in traditional Chinese medicines and health care products.Method:An Agilent ZORBAX-Eclipse XDB-18(4.6 mm×250 mm,5 μm)column was adopted with a mobile phase composed of 0.02 mol·L-1KH2PO4(containing 2 mmol·L-1 SDS)-acetonitrile(62.5∶37.5,adjusted to pH 6.5 using 1 mol·L-1 NaOH solution)at a flow rate of 1.0 mL·min-1.Detection was carried out at 230 nm,and the column temperature was 30 ℃.Results:The 9 type of chemical anti-diabetic agents were separated well under the selected chromatographic conditions.The linear regression equation for each component was obtained,and the correlation coefficient(r)were better than 0.9992.The limits of detection were 5.9-43.5 μg·L-1.Conclusion:The method can be used for the detection of the anti-diabetic agents in traditional Chinese medicines and health care products,and it is rapid,convenient,economic and specific.
