淫羊藿含药血清对绝经后骨质疏松症大鼠MSCs成脂分化干预作用研究

The research on the effect of Yinyanghuo-containing serum on MSCs induced adipogenesis in postmenopausal osteoporosis rat

目的:探讨淫羊藿含药血清对绝经后骨质疏松症大鼠MSCs成脂分化的干预作用。方法:SPF级SD雌性大鼠,共15只。无菌条件下分离培养骨髓间充质干细胞,传至3代。将所获的细胞随机分为空白组、成脂诱导组、成脂诱导西药组、成脂诱导中药高剂量组、成脂诱导中药中剂量组、成脂诱导中药低剂量组共6组,每组8孔。通过淫羊藿高、中、低剂量含药血清以及尼尔雌醇含药血清配合成脂诱导剂分别对MSCs进行干预,并与空白组、成脂诱导剂组进行对比;观察各组干预后7,14d成脂分化情况。结果:诱导第7d,与空白组比较,中药各组油红染色均呈显著性差异(P<0.05);成脂诱导组呈极显著性差异(P<0.01);与成脂诱导组比较,成脂诱导西药组油红染色有极显著性差异(P<0.01),但成脂诱导中药组呈显著性差异(P<0.05)。诱导第14d,各组与空白组比较均呈显著性或极显著性差异(P<0.05,P<0.01);与成脂诱导组比较,成脂诱导中药高剂量组油红染色呈显著性差异(P<0.05),成脂诱导西药组有极显著性差异(P<0.01)。结论:成脂诱导中药高剂量组及成脂诱导西药组均能抑制MSCs成脂分化,但短时间内高剂量淫羊藿含药血清功效略逊于西药尼尔雌醇。

Objective：To observe the effect of Yinyanghuo-containing serum on MSCs induced adipogenesis in postmenopausal osteoporosis rat.Method：15 SPF SD rats were divided into 6 groups at random as control group,adipogenic group,adipogenesis and western medicine group,adipogenesis and high does Chinese medicine group,adipogenesis and normal does Chinese medicine group,adipogenesis and low does Chinese medicine group.After 7 and 14 days,to observe the different results of each group,to draw a conclusion whether the MSCs can be induced into adipogenesis or the effect is inhibited by the medicine.Result：Compared with the control group,adipogenic group and every Chinese medicine group are significantly higher（P0.05,P0.01）.while compared with adipogenic group,adipogenesis and western medicine group is much significantly lower than the other groups（P0.01）.After being induced 14 days,both adipogenesis and western medicine group and adipogenesis and Chinese medicine groups can inhibite？MSCs adipogenic induction（P0.05,P0.01）,but the adipogenesis and western medicine group get the better effect.Conclusion：Both adipogenesis and western medicine group and adipogenesis and Chinese medicine groups can inhibite MSCs adipogenic induction.But Yinyanghuo-containing serum＇s effect is not as good as western medicine in short time.
