摘要
骨唾液酸蛋白(bone sialoprotein,BSP)是一种高度糖基化、磷酸化、硫酸化的多功能非胶原蛋白,主要分布于矿化相关组织,多种恶性肿瘤中也有异常表达。BSP基因转录调控序列位于5’端侧翼区,由启动子、增强子、沉默子、糖皮质激素应答元件等组成。基因转录的顺式作用元件位于启动子区,顺式作用元件及其结合的转录因子有生物种类特异性,一些转录因子以不同的亲和力与其部分潜在结合位点结合,并相互作用协同调控转录。激素、成纤维细胞生长因子-2(fibroblast growth factor 2,FGF-2)、胰岛素样生长因子-1(insulin-like growth factor 1,IGF-1)、α-肿瘤坏死因子(tumor necrosis factor-α,TNF-α)等通过特定的信号通路参与对BSP基因的转录调控,在BSP基因启动子上有它们的应答元件。
Bone sialoprotein(BSP) is highly glycosylated,phosphorylated and sulfated multifunctional noncollagenous protein that is essentially unique to mineralizing connective tissues,also expressed in a variety of malignant tumors.The transcriptional regulation sequences located in the 5' flanking region of the BSP gene encompass promoter,enhancer,silencer,and a glucocor-ticoid response element.The control effects of the cis-acting elements of the BSP promoter and binding proteins-transcription factors depend on the specificities of cells.Some transcription factors bind to the potential sites of the gene promoter with different affinity and also interact with each other to synergistically regulate BSP transcription.Many factors participate in transcriptional regulation of BSP gene in which there are their response elements via certain signaling pathways,including hormones,FGF-2,IGF-1,TNF-α,etc.
