脑内皮细胞趋化因子受体5参与β淀粉样蛋白引起的T细胞跨内皮迁移 被引量:1

摘要 目的利用血-脑屏障的体外模型,分析脑内皮细胞上的趋化因子受体5(CCR5)在β淀粉样蛋白(Aβ)引起的T细胞跨内皮迁移过程中的作用。方法培养人脑微血管内皮细胞(HBMEC),建立体外血-脑屏障模型,分析T淋巴细胞跨内皮迁移的能力。结果 Aβ能够促进T淋巴细胞穿过体外血-脑屏障模型,HBMEC中CCR5过表达能够促进T淋巴细胞穿过体外血脑屏障模型,HBMEC中CCR5缺失突变能够抑制T淋巴细胞穿过体外血-脑屏障模型。结论 HBMEC中CCR5参与Aβ引起的T淋巴细胞的跨内皮迁移。 Objective To investigate the function of chemokine receptor 5(CCR5)in human brain microvascular endothelial cells(HBMEC)during T lymphocytes transendothelial migration induced by amyloid beta(Aβ).Methods The in vitro model of blood-brain barrier was established with cultured HBMEC monolayer.The ability of T lymphocytes transendothelial migration induced by Aβ was analyzed by Transwell and Millicell-ERS endothelia volt-ohmmeter.Results Aβ could promote T lymphocytes to migrate through the HBMEC monolayer.Over-expression of CCR5 in HBMEC promoted T lymphocytes to migrate through HBMEC monolayer.Expression of CCR5 mutant in HBMEC inhibited the transendothelial migration of T lymphocytes.Conclusion CCR5 of HBMEC is involved in T lymphocytes transendothelial migration induced by Aβ.
