三苯氧胺对人脑胶质瘤细胞内游离钙离子水平的影响 被引量:2

Effects of tamoxifen on the cytosolic free calcium level in human SHG-44 cells

摘要 以不同浓度的三苯氧胺(TAM)处理SHG-44细胞后,用流式细胞仪检测细胞凋亡率和激光共聚焦显微镜测定细胞内游离Ca2+水平,探讨TAM对人脑胶质瘤细胞内游离钙离子水平的影响。结果表明:当细胞外有Ca2+时,TAM使细胞内Ca2+浓度迅速升高并呈剂量依赖性关系,其浓度能维持在较高水平。当细胞外无Ca2+时,TAM呈剂量依赖性地使胞内Ca2+浓度缓慢升高,其升高的幅度和速度低于细胞外有Ca2+组。说明TAM能促进SHG-44细胞胞外Ca2+内流和胞内钙库释放Ca2+,提高细胞内游离Ca2+浓度;TAM诱导的细胞凋亡与提高细胞内游离Ca2+浓度有关。 To investigate the effects of tamoxifen on the cytosolic free calcium level, the apoptotic rate was measured by flow cytometry assay and the cytosolic free calcium level was determined by laser confocal microscope assay after treating human glioma cell SHG-44 with various concentration of tamoxifen. Result showed that, in medium containing extracellular Ca^2+ , tamoxifen promptly increased cytosolic free calcium level in a dose-dependent manner, and afterwards it remained at a high level. Although tamoxifen also increased cytosolic free calcium level in a dose-dependent manner without extracellular calcium, it showed much slower and less. In conclusion, tamoxifen is likely to increase cytosolic calcium by releasing intracellular calcium and/or causing extracellular calcium influx. Apoptosis induced by tamoxifen may be related to its ability to increase the cytosolic free calcium level in human glioma cell SHG-44.
